Friedrich et al. have reported that the analog with 1,1-dimethyl-l-amino-2-ethanol in place of 1-amino-2-propanol is inactive as a coenzyme in the diol-dehydrase system, although it is bound to the enzyme as tightly as is cyanocobalamin (31). 

This vitamin possesses the most complex structure of any of the vitamins and is unique in that it has a metallic element, cobalt, in the molecule (Figure 9-19). The molecule is a coordination complex built around a central tervalent cobalt atom and consists of two major parts—a complex cyclic stmcture that closely resembles the porphyrins and a nucleotide-like portion, 5,6-dimethyl-l-(a-D-ribofuranosyl) benzimidazole-3 -phosphate. The phosphate of the nucleotide is esterified with 1-amino-2-propanol this, in turn, is joined by means of an amide bond with the propionic acid side chain of the large cyclic stmcture. A variety of alkyl amines B, including 1-propylamine, ethylene diamine, 1,3-diaminopropane, and (R)-1 -amino-2-propanol have been used as reactants. The guest exchange kinetic results are reported in Table 13. The presence of more than one reacting [/3-CD-H-A]4" structure is observed with A = DOPA and penicillamine. The results have been rationalized in terms of specific interactions in the relevant inclusion complexes which determine their structure and relative stability.
