Phosphorylation triggers a second series of events. Phosphorylated amino acids residues act as docking sites for a number of intracellular proteins. These proteins contain their own amino acid motifs that recognize and bind to one or another of the phosphorylated amino acids. Some of these proteins have additional domains that now become activated and bind to other signaling molecules. A complex of proteins is thus assembled at the surface of the cell and specific signaling... [Pg.73]

Mechanism-based irreversible inhibitors and mutational analysis with OSCs have shown that the highly conserved amino acid motif DCTAE is required for substrate binding [27,46,57-59] (Table 3), and the conserved aspartate residue within this motif (D456) has been implicated as the likely electrophilic activator in the generation of the protosteryl cation for LS [57, 58, 60]. Similar experiments indicate that two aspartate residues at the homologous position of the... [Pg.40]

Tertiary alcohols are very unreactive toward hydrolases. There are, however, exceptions. Some enzymes have a certain amino acid motif located in the oxyanion binding pocket that allows the docking of space-demanding alcohols such as tertiary ones into the acylated enzyme [104]. One such hydrolase is Candida antarctica lipase A (CALA), which has been found to catalyze the acylation of the tertiary 2-phenyl-3-butyn-2-ol rac-110 by vinyl acetate in organic solvents. Thus efficient resolution of 110 was achieved in isooctane at room temperature (Scheme 4.34) [105]. [Pg.100]

The evolution of protein structures shows conservation in recognition features for common binding sites. For example, the adenosine triphosphate (ATP)/ guanosine triphosphate (GTP) binding site contains an amino acid motif A (P-... [Pg.31]

Core N-glycan structure. This consists of a core pentasaccharide composed of a trimannosyl group attached to two Glc-NAc residues. The structure is attached to an asparagine (Asn) residue in a sequon which is an amino acid motif of three amino acids. X is any amino acid (except Pro) that is positioned between Asn and either serine or threonine. [Pg.130]

Zonulin, an intestinal Zot analogue sharing an 8-amino acid motif with Zot (Di Pierro et al. 2001), was shown in a primate model to reversibly open intestinal tight junctions after engagement to the same receptor activated by Zot (Wang et al. 2000) The physiological role of Zonulin remains unclear but it... [Pg.93]
