The discovery and exploitation of enzymes in aldoxime-nitrile pathway nitrile hydratase, amidase, nitrilase, aldoxime dehydratase, etc., are shown along with the use of methodologies, such as organic chemistry, microbial screening by enrichment and acclimation culture techniques, enzyme purification, gene cloning, molecular screening by polymerase chain reaction (PCR). [Pg.129]

Smdies in this area led to the proposal that the aldoxime-nitrile pathway, which is implemented in synthesis of drugs and fine chemicals, occurs as a natural enzymic pathway. It is of interest that the enzyme responsible for bacterial conversion of Af-hydroxy-L-phenylalanine to phenacetylaldoxime, an oxidative decarboxylation reaction, lacks heme or flavin groups which are found in plant or human enzymes that catalyze the same reaction. Its dependency on pyridoxal phosphate raised the possibility that similar systems may also be present in plants . [Pg.637]

The NHase responsible for aldoxime metabolism from the i -pyridine-3-aldoxime-degrading bacterium, Rhodococcus sp. strain YH3-3, was purified and characterized. Addition of cobalt ion was necessary for the formation of enzyme. The native enzyme had a Mr of 130000 and consisted of two subunits (a-subunit, 27 100 (3-subunit, 34500). The enzyme contained approximately 2 mol cobalt per mol enzyme. The enzyme had a wide substrate specificity it acted on aliphatic saturated and unsaturated as well as aromatic nitriles. The N-terminus of the (3-subunit showed good sequence similarities with those of other NHases. Thus, this NHase is part of the metabolic pathway for aldoximes in microorganisms. [Pg.136]

The oxidation of aromatic aldoximes with ceric ammonium nitrate produces nitrile oxides which undergo subsequent cycloaddition to nitriles to produce 1,2,4-oxadiazoles (Equation 47) <1997PJC1093>. The anodic oxidation of aromatic aldoximes in the presence of acetonitrile has been reported to give low yields of either 3-aryl-5-methyl-1,2,4-oxadiazoles (2-25%) or 3,5-bis-aryl-l,2,4-oxadiazoles (6-28%), although the synthetic utility of this route is limited by competitive deoximation to the carbonyl being the major reaction pathway <1997MI3509>. [Pg.280]
