In conclusion, it is likely that computational approaches for metabolism prediction will continue to be developed and integrated with other algorithms for pharmaceutical research and development, which may in turn ultimately aid in their more widespread use in both industry and academia. Such models may already be having some impact when integrated with bioanalytical approaches to narrow the search for possible metabolites that are experimentally observed. Software that can be updated by the user as new metabolism information becomes available would also be of further potential value. The held of metabolism prediction has therefore advanced rapidly over the past decade, and it will be important to maintain this momentum in the future as the hndings from crystal structures for many discrete metabolic enzymes are integrated with the diverse types of computational models already derived. 

Taxonomically close to the Annonaceae, the Lauraceae family abounds with apor-phinoid alkaloids. A remarkable advance in the search for topoisomerase inhibitors from Lauraceae has been provided by Woo et al. (6). Using DNA-unwinding assay and structural modeling, they showed that dicentrine can attain a relatively planar conformation and molecular bulk which allow it to occupy the active site of topoisomerase II which becomes inactive. The requirement of a suboptimal conformation to achieve DNA binding appears to make dicentrine less potent against topoisomerase II than the... 

A detailed treatment of the evolution of the genetic code requires modelling physical components of the translational process this includes the dynamic processes of the tRNAs and the aminoacyl-tRNA synthetases (Vetsigian et al., 2006). Thus, in spite of considerable advances in the search for the roots of the genetic code, there is still much to do ... 

Progress in these areas will require a number of new supporting tools that can effectively handle and solve a variety of mathematical models involving thousands and millions of variables. These supporting tools in turn will require that chemical engineers become acquainted with new advances in numerical analysis, mathematical programming, and local search techniques.
