5HTia receptor

Novel agonist 5HTia receptor position emission tomography (PET) ligands derived form l,2,4-triazine-3,5-dione 19 have been synthesized and evaluated in vivo <06JMC125 06BMCL2101>.

Buspirone is a partial agonist at 5HTia receptors St John s wort is a non-selective inhibitor of 5HT reuptake and also upregulates postsynaptic 5HTia and 5HT2a receptors it therefore causes overstimulation of 5HTia receptors, leading to the serotonin syndrome.

Buspirone is used as an anxiolytic and response to treatment may take two to four weeks. This drug is thought to work as an agonist on 5HTia receptors. These are pre-synaptic receptors that limit the release of serotonin. Buspirone does not have the dependence potential of the benzodiazepines. 

Urapidil is an alpha-blocker and has a central sympatholytic effect mediated by stimulation of central 5HTia receptors. The authors of a review have suggested that there is a lower incidence of tachycardia and orthostatic hypotension with urapidil than with other a-adrenoceptor antagonists and no apparent increased risk of heart failure [101 ].
