Figure 7 Denaturing gradient gel (35-70%) of PCR-amplified 16S rDNA fragments from untreated reprocessed mine tailings (RT), unlimed RT columns (replicates A and B), limed RT columns (replicates A and B), and C. leidyi, S. yanoikuyae, and R. loti-like isolates cultivated from the taihngs after column treatments. To resolve the S. yanoikuyae and R. loti bands in the hmed RT column (replication A), the sample was run with a narrower (40-60%) denaturing gradient (inset). Comigration of isolate bands with bands from limed RT columns is indicated with dashed hues (From Ref. 58.)... Figure 14 Denaturing gradient gel (35-70%) of PCR-amplifled 16S rDNA fragments from nontreated Van Dyke soil (Madison River basin) and from column experiments using this soil under saturated flow [As(V) influent], unsaturated flow [As(III) influent], and unsaturated flow [As(V) influent] conditions. In addition, the banding patterns for two of the isolates cultivated from these treatments (Psuedomonas fluorescens and Agrobacterium tumefaciens) are shown to comigrate with bands in the environmental samples as indicated with dashed lines. FISH, terminal restriction-fragment length polymorphism analysis (rRNA-based molecular techniques) and comparative 16S rDNA analysis... [Pg.17]

However, composition of biofilm samples were similar (according to restriction analysis of the 16S rDNA, at this level of differentation) and all three investigated bacterial groups were detected by PCR. Difference was only observed in the intensity of restriction fragments, which may suggest the dominance of different species around the various sampling points. The Article 5 of 98/83/EC Directive allows Member States to set values for additional parameters not included in Annex I where the protection of human health within its national territory or part of requires it so. In our opinion this kind of examinations may be very useful in evaluation of disinfection efficiency. [Pg.506]
