Zygosaccharomyces rouxii

On the synthetic route to a novel class of orally active 2,3-benzodiazepines, scientists at Lilly have described a yeast reduction using Zygosaccharomyces rouxii (ATCC 14462) (Scheme 19.5).76-78 This intermediate is used in the synthesis of a novel class of benzodiazepines such as LY-300164 (1). The choice of this yeast strain came from a screen of numerous microbes. The product was isolated in >95% yield and >99.9% ee by use of an adsorbant polymeric resin.76-78... [Pg.364]

Fig. 3.51). The chiral alcohol is a key intermediate in the synthesis of an anticonvulsant drug. The yeast Zygosaccharomyces rouxii employed for this process is hampered by substrate and product toxicity at levels of >6gL-1. This was solved by using in situ adsorption on Amberlite XAD-7. In this way in situ product removal could be achieved. At the end of the process (8-12 h) 75-80 g of the alcohol is found on the resin and about 2 g L-1 remains in the aqueous phase. Finally an isolated yield of 85-90% could be obtained with an ee>99.9%. Fig. 3.51). The chiral alcohol is a key intermediate in the synthesis of an anticonvulsant drug. Talampanol (LY300164) is a drug used to treat epilepsy and neurodegen-erative diseases. In an optimized procedure, the first step in its production is enzymatic in nature (Figure 10.4). Zygosaccharomyces rouxii is used to perform a biocatalytic reduction and excellent yields and enantioselectivities have been... [Pg.207]

In an effort to reduce benzylmethylketone 6 Anderson et al. identified that the yeast Zygosaccharomyces rouxii could catalyze this transformation efficiently at low substrate concentration (Scheme 4.10). Since the substrate and product were both toxic to Z. rouxii, polymeric hydrophobic resins were used to both supply substrate and remove the product from the reaction mixture as it formed. This approach allowed the reaction concentration to be increased from 2 to 40 g L . The... [Pg.106]

A nonionic cross-linked polymethacrylate resin (Am-berlite XAD7) was used in the reduction of the ketone (5.34) by an aqueous suspension of Zygosaccharomyces rouxii cells.145 Adsorption by the resin kept the concentrations of the substrate and product below levels that would be lethal to the cells, yet allowed enough to be in solution for the reduction to take place in 96% yield with more than 99.9% enantiomeric excess (often abbreviated %ee). [Pg.118]

In Zygosaccharomyces rouxii, one of the few salt-tolerant yeasts, the amount of free ergosterol in the membrane of cells grown in 15% NaCl increased 2.9-fold over that in control cells. This increase could have contributed (together with the decrease of fatty acid unsaturation) to the observed loss of membrane fluidity. Such a modification would facilitate... [Pg.1033]
