X- Phosphate Phosphate Phosphoric acid Phosphates, alcohols... 

Digoxigenin (Dig) Detection Kit (Boehringer-Mannheim), containing anti-Dig AP conjugate, X-phosphate, and 4-nitro blue tetrazolium chloride (NBT). 

To the second universal add 22 pL of NBT + 17 pL of X-phosphate 5-bromo-4-chloro-3-indolyl-phosphate (BCIP) for both biotin and dig probes. Wrap both universal tubes in tinfoil to keep out the light.

BCIP (= X-phosphate 5-bromo-4-chloro-3-indolyl phosphate) 1.75 mg is dissolved in 35 pi of 100% DMF and added dropwise with gentle mixing into the NBT solution. Final concentration 0.33 mg/ml NBT and 0.17 mg/ ml BCIP.

The combination of a reduced plasma calcium concentration (due to vitamin D deficiency) and a reduced plasma phosphate concentration (due to secondary hyperparathyroidism) produces a calcium X phosphate product below the threshold for calcification of new bone.
