For the accumulated costs and resources devoted to the development of a new chemical entity (NCE) or new molecular entity (NME) to make sense financially, the commercial potential of the compound must be evaluated in a rigorous manner. Compounds whose expected financial performance does not warrant these high investment costs must be abandoned or out-licensed as soon as possible so as to direct resources toward more profitable endeavors. By operating effectively, a well-designed drug discovery and development process can focus its efforts to operate efficiently on the compounds that will maximize cash flow to the pharmaceutical firm. [Pg.619]

Our previous work with PPNVP and GOx-PPNVP/PEUU (6,12,25), which focused on characterization of the surface modified thin films, has demonstrated that a reasonable amount of control can be exerted over the chemistry of plasma modification. The techniques used here to attach active GOx to PEUU may be applied to a wide variety of biomolecules and a wide variety of organic and inorganic substrates. Incorporation of GOx-PPNVP/PEUU into the thin-layer cells extended the potential applicability of GOx-PPNVP/PEUU and similar materials to specific practical applications such as sensing devices. The monocyclic substrates are well suited for discussion of relative stereocontrol. In a recently reported study employing cis- and rrar -disubstituted substrates (9) and (10). high stereoselectivity could be obtained when gallium trichloride was used as the Lewis acid (Scheme 12). In these reactions new stereocenters can be created at C(3) and C(3a). In the cis series both tautomeric products were formed as single stereoisomers while the trans series (10) produced a mixture of the tetrasubstituted isomers. A post facto epimerization was suggested as the cause for the lower selectivity.
