Verification Methods for Robustness in RP-HPLC

During the daily routine in an HPLC laboratory, problems often arise that may result from a lack of robustness of the RP-HPLC methods used. [Pg.643]

The robustness of an RP-HPLC method can generally be established in two different ways, either during a systematic method development [1, 6,10,11] or by the application of a statistical experimental design [2-5,13, 15-36]. [Pg.643]

A good approach is the systematic change of the important chromatographic parameters and the measurement of the resulting effects on the separation. This procedure applies to both ways of establishing robustness. [Pg.643]

Testing Robustness in Analytical RP-HPLC by Means of Systematic Method Development [Pg.643]

Computer-facilitated HPLC method development using a simulation program (e.g., DryLab, ACD LC GC Simulator) can be very useful for the investigation of the influence of the respective chromatographic parameters on the separation and consequently on the robustness of a given RP-HPLC method. [Pg.643]
