The information obtained by this preliminary analysis can be used not only to set priorities for the various possible Diels-Alder disconnections, but also to pinpoint obstacles to transform application. Recognition of such obstacles can also serve to guide the search for specific retrosynthetic sequences or for the highest priority disconnections. At this point it is likely that... [Pg.18]

Aldehyde 198 served as a key intermediate in a synthesis of the alkaloid ajmaline. The. Mannich aminomethylation transform triggers disconnection of two bonds in 198 to form dialdehyde 199, which by connective transform application can be converted to cyclopentene 200.58,59 The reduction in functional group reactivity and in structural complexity are both apparent by comparison of 198 and 200. [Pg.73]

Sato et al. carried out detailed studies on the possibilities of transformation of tetrazolo[l,5-tf]pyrazines 54 to 2-aminopyrazines 56 < 1994S931 >. These authors found that the generally used methods for this conversion fail because the starting compound exists in the stable bicyclic form 54, whereas partial formation of the azide valence bond isomer 55 would be necessary for the success of the transformation. Application of special reaction conditions succeeded, however hydrogenation over palladium catalyst in the presence of ammonium hydroxide or treatment with stannous chloride in a mixture of methanol and hydrochloric acid solved this problem. Thus, a great number of derivatives of 54 was reduced to the corresponding 2-aminopyrazine 56 in medium to high yields (45-100%). [Pg.826]
