Transfection competence

Comment It is critical to remove excess cap analogue from in vitro transcribed transcripts prior to transfection, because the cap analogue will compete with transcripts for the cellular translational machinery. Also, even after DNase treatment, the RNA sample may still contain traces of functional pDNA, which may interfere with subsequent detection by RT-PCR. Furthermore, plasmids containing a mammalian promoter may even give rise to de novo transcription in transfected cells. [Pg.123]

The DNA or cDNA library is then introduced into a preparation of bacterial host cells. Usually, the first host selected is a laboratory strain of E. coli which has been grown and pretreated with inorganic salts to make uptake of DNA easier. The ability to take up foreign DNA is called competence, cells which have been specially prepared for the purpose are called competent cells. Other methods to transfer DNA into cells include electroporation (application of an external electric field to permeabUize the cell wall), transfection (where a recombinant bacterial virus is used to transfer the DNA to the target cell) or ballistic methods (by using DNA-coated particle projectiles). The last method has been used to introduce foreign DNA into plant cells and mammalian cells. [Pg.101]

OptiMEM I reduced-serum medium is widely used for complex formation. Other serum-free media can also be used as long as they are free of polyanions such as heparin or dextran-sulfate, which compete with DNA or nucleic acids to bind to the transfection reagent. [Pg.46]

Anandamide parallels d9-THC in competing with the binding of [3H]HU-243 (6) to the brain cannabinoid receptor [33]. However, under most experimental conditions an amidase blocker has to be used in order to prevent anandamide hydrolysis during the binding experiments [38], Using neuroblastoma cells that express this receptor naturally, as well as cultured cell lines transfected with this receptor, it was shown that, like d9-THC, anandamide inhibits A-type voltage-dependent calcium channels [39] and adenylate cyclase [40, 41],... [Pg.205]

Two standard methods used to study protein function are transfection of the protein of interest or knock down of the target protein. Transfected proteins may compete with endogenous... [Pg.976]
