Gronowitz et observed a similar isomerization of (2-pyrrolyl-thio)acetic acid to (3-pyrrolylthio)acetic acid by polyphosphoric acid followed by cyclization to 2,3-dihydrothieno[3,2-h]pyrrol-3-one. 

The first syntheses of 1,2-cfs-thioglycoses (a-D-gluco- and )3-D-manno- derivatives) have been achieved by the reaction in acetone of alkyl or benzyl xanthate or potassium thioacetate with the corresponding l,2-tra s-glycosyl halides [13]. More recently, tetra-O-acetyl-l-S-acetyl-l-thio-a-D-glucopyranose (10a) (Scheme 3) has been obtained i) by reaction of -acetochloroglucose (9 a) with either potassium thioacetate in HMPA or the tetrabutylammonium salt of thio-acetic acid in toluene [14] ii) by peroxide-induced addition of thioacetic acid to the pseudo-glucal (11) [15]. 

Reactions of 2-[l-(thietan-3-yl)-benzimidazolyl-2-thio]acetic acid 43a and 2-[T(l,l-dioxothietan-3-yl)-benzimida-zolyl-2-thio]acetic acid 43b with bases (amino alcohols or sodium hydroxide) led to the corresponding salts 44a-h. By treatment of the ethyl esters 45a and 45b, obtained by the reaction of acids 43a and 43b with ethanol in the presence of sulfuric acid, with hydrazines, (benzimidazolyl-2-thio)acetic acid hydrazides 46a and 46b were synthesized and subsequently converted into the corresponding dihydrochlorides 47a and 47b (Scheme 16). Salts 44a-h and 47a and 47b were tested as immunotropic compounds <2001PCJ11>. 

Cyclization of [(l-isoquinolinylmethyl)thio]acetic acids (249) on the action of DDQ or Ac20 afforded [l,4]thiazino[3,4- ]isoquinolin-4-ones 250 (81GEP3023717). l,2-Dihydro[l,4]thiazino[4,3-a]quinolin-l-ones were obtained from (2-quinolyl)methylmercaptoacetic acids by treatment with Ac2Q in pyridine (73IJC1051).
