Testing implementation approach

The hardware and software used to implement LIMS systems must be vahdated. Computers and networks need to be examined for potential impact of component failure on LIMS data. Security concerns regarding control of access to LIMS information must be addressed. Software, operating systems, and database management systems used in the implementation of LIMS systems must be vahdated to protect against data cormption and loss. Mechanisms for fault-tolerant operation and LIMS data backup and restoration should be documented and tested. One approach to vahdation of LIMS hardware and software is to choose vendors whose products are precertified however, the ultimate responsibihty for vahdation remains with the user. Vahdating the LIMS system s operation involves a substantial amount of work, and an adequate vahdation infrastmcture is a prerequisite for the constmction of a dependable and flexible LIMS system. 

The application of density functional theory to isolated, organic molecules is still in relative infancy compared with the use of Hartree-Fock methods. There continues to be a steady stream of publications designed to assess the performance of the various approaches to DFT. As we have discussed there is a plethora of ways in which density functional theory can be implemented with different functional forms for the basis set (Gaussians, Slater type orbitals, or numerical), different expressions for the exchange and correlation contributions within the local density approximation, different expressions for the gradient corrections and different ways to solve the Kohn-Sham equations to achieve self-consistency. This contrasts with the situation for Hartree-Fock calculations, wlrich mostly use one of a series of tried and tested Gaussian basis sets and where there is a substantial body of literature to help choose the most appropriate method for incorporating post-Hartree-Fock methods, should that be desired. 

Another group of methods for testing 3D models that implicitly take into account many of the criteria listed above involve 3D profiles and statistical potentials [87,216]. These methods evaluate the environment of each residue in a model with respect to the expected environment as found in the high resolution X-ray structures. Programs implementing this approach include VERIFY3D [216], PROSA [217], HARMONY [218], and ANOLEA [120]. 

As part of developing the PSM implementation plan (Chapter 5), you and the team identified benefits unique to the approach you selected, using them to help win management s approval. For example, your plan may focus on priority elements because the assessment you conducted suggests that this method will yield the greatest overall improvement in safety performance. And, as part of the pilot test described in this chapter, you focused on facility-specific benefits to enlist the support of local management and staff.
