Tablet formulation caffeine

Tablet Formulations (Immediate Release). Two papers in the mid-1990s reported the earliest studies on immediate release tablets. In the first, tablet formulations of hydrochlorothiazide [33] were modeled in an attempt to maximize tablet strength and select the best lubricant. In the other, a tablet formulation of caffeine was modeled [34] to relate both formulation and processing variables with granule and tablet properties.

From 1982 through 1985, few NIR analyses of dosage forms were published. Since 1986, there have been many articles. The first was a 1986 paper by Ciurczak and Maldacker [33] using NIR for tablet formulation blends, examining the use of spectral subtraction, spectral reconstruction, and discriminant analysis. Blends were prepared where actives—aspirin (ASA), butalbital (BUT), and caffeine (CAF)—were omitted from the formulation or varied over a range from 90 to 110% of label strength. 

A 41-year-old woman presented with pain and pallor in the leg and a sensation of coolness exacerbated by exercise (34). For many years she had been taking a formulation containing ergotamine 1 mg plus caffeine 100 mg, at a dose of one or two tablets daily, for both prophylaxis and treatment of migraine. For 7 days she had also taken clarithromycin (dose is not stated) for a chest infection. Her legs were cool and cyanosed, with no palpable popliteal or foot pulses and an ankle-brachial index of only 0.6 (normal >0.8). 

As a pharmaceutical excipient, vanillin is used in tablets, solutions (0.01-0.02 % w/v), syrups, and powders to mask the unpleasant taste and odor characteristics of certain formulations, such as caffeine tablets and polythiazide tablets. It is similarly used in film coatings to mask the taste and odor of vitamin tablets.
