Quinolones, (I), and lH-imidazo[4,5-b]pyridine derivatives, (n), effective as methionyl t-RNA synthetase inhibitors were previously prepared by the author (2,3), respectively. 

Aminopiperidine quinoline derivatives, (III), (IV), and (V), prepared by Davies (4,5) and Markwell (6), respectively, were effective as methionyl t-RNA synthetase inhibitors against S. aureus Oxford, S. aureus WCUH29, S. pneumoniae 1629, S. pneumoniae N1387, and S. pneumoniae ERY 2.

The instrument is tunable the other oblique-cut crystal available is 5° Ge(lll) (Lemonnier et al 1978). This was the first oscillation camera film data collection service at a synchrotron. One of the earliest protein crystal data sets to be collected was from single crystals of tyrosyl t-RNA synthetase (Monteilhet, Fourme and Blow 1978). Many data sets have now been collected (summaries of which are given in Fourme (1978, 1979), Fourme and Kahn (1981), Kahn et al (1982a), Bartunik, Fourme and Phillips (1982) and Wilson et al (1983)). 

On DCI, Lemonnier et al (1978) established a focussing monochromator camera in 1976 (section 5.6.3.1) on which oscillation camera data sets were collected (e.g. from single crystals of tyrosyl t-RNA synthetase (Monteilhet et al 1978) or resolution and radiation damage tests were conducted (e.g. 6-phosphogluconate dehydrogenase (Helliwell 1979)). 

An inhibitor of seryl t-RNA synthetase, SB-217452, isolated from a Strepto-myces species, has been identified as the 4 -thionucleoside 177. The nucleoside moiety seems to be identical to one obtained as an enzymic cleavage product of albomycin 62 (Vol. 18, p. 182). ... 

Synthesis using Aminoacyl t-RNA Synthetase. Other interesting synthetic reactions using ATP are the use of aminoacyl t-RNA synthetase (ARS, E.C. sub-class 6.1.1) to synthesize peptides (9-72) and for the construction of the fascinating biologically-active nucleotide, diadenosine polyphosphate (Ap4A) (13-14). ARS isolated from mesophiles is quite unstable and is usually handled on ice. ARS from B. stearothermophilus, on the other hand, is quite stable and is a useful catalyst for bioreactors. 

It is worth mentioning that dye-ligand affinity chromatography has found application within a relatively short time in the isolation and purification of a series of dehydrogenases, phosphotransferases, t-RNA synthetases, restriction nucleases, initiation factors of protein biosynthesis, a-fetoprotein, clotting factors II and X and complement factors. 

The dependency of bacterial RNA synthesis on protein synthesis which has been called "stringent control" has been demonstrated to take place under conditions such as aminoacid-starvation, nitrogen starvation, phosphate and Mg" starvation, energy deprivation and in strains mutated in amino-acyl-t-RNA synthetases. In all these conditions the synthesis of r-RNA and t-RNA is inhibited as far as the m-RNAs are concerned, some m-RNAs are inhibited, whereas the synthesis of others is enhanced" ". ... 

Catalytic residues present in motif 2 and motif 3 of Class II aminoacyl t-RNA synthetases (EC - E. coli SC - S. cerevisiae HA - Hamster TT - Thermus thermopilus)... 

The geometry of superimposed residues (Fig. 4) is substantially different from analogous model obtained for Class I t-RNA synthetases 40]. It indicates that the electrostatic field enhancing the rate of ATP aminoacylation... 

The approach has been employed to determine the amino acid sequence in E, coli alanine t-RNA synthetase. After partial hydrolysis, the peptide mixtures were converted to their methyl esters, N-trifluoroacetylated, reduced with B2D6 and converted to their O-TMS derivatives (9). These compounds possess good GC properties and yield simple, yet informative fragmentations. A variety of derivatives have been explored for sensitive sequence analysis of small peptides and a new permethylation procedure has been described for trifluoroacetylated peptides at the 2 —10 nmol level. Mixtures of these derivatives, ideally of di- to hexa-peptides are readily analyzed by GC/MS 66).
