The nature of OA interaction with SC lipids in vivo was of significant interest, particularly in light of the IR results above. Reflectance IR spectroscopy in conjunction with the use of a deuterated probe once again proved to be a valuable approach for the noninvasive evaluation of this enhancer in humans [153]. Prior to treatment, test sites on the inner ventral forearm of volunteers were cleansed with water, after which the subject remained at constant temperature and relative humidity, while three pretreatment spectra were collected. The experimental site on one arm was treated with a 5% v/v solution of perdeuterated oleic acid ( H-OA) in ethanol, while the control site, on the contralateral arm, was treated with ethanol alone. Both formulations were applied under occlusion for 16 hours posttreatment, the sites were swabbed clean with ethanol and then air-exposed for 2 hours to allow the occluded skin to dry. An ATR-IR spectmm of the dosed site was then obtained. This site was then tape stripped once and a second spectral examination was made. This sequential tape stripping and spectral acquisition was repeated -20 times in order to obtain an incremental spectral profile as a function of SC depth (defined by the cumulative weight of SC removed with tape stripping). It may be desirable to first clean the pipeline with a polyurethane foam swab. This material can be purchased commercially either in specific cut sizes, or in bulk, which can be cut to the desired size. Swabs will effectively remove soft scales and loose material. Fill a cuvette about three-fourths full with distilled water and dry its outside with a tissue. To calibrate the colorimeter, place the cuvette in the colorimeter and close the lid. Turn the wavelength knob to 0%T. Press TRIGGER on the CBL and enter 0 into the calculator. Turn the wavelength knob to Red (635 nm). Press TRIGGER on the CBL and enter 100 into the calculator. Leave the colorimeter set on Red for the rest of the lab. Remove the cuvette from the colorimeter. Empty the distilled water from the cuvette. Clean the cuvette with a cotton swab and fill it about three-fourths full with the unknown dye solution. Place the cuvette in the colorimeter and close the lid. From the MAIN MENU, select COLLECT DATA (do not select SET UP PROBES as this will erase your data lists). Select MONITOR INPUT from the DATA COLLECTION MENU. When all the commodities and supplies have been selected, the stability of the analyte in the rinse or swab solution must be determined. This should be done at more than one concentration and if analyte determination is noted, refrigeration or freezing of the solutions should be evaluated, to extend the analysts testing window. Degraded out-of-date samples will give lower numbers than true numbers and a false sense of cleaning effectiveness. [Pg.407]

The last step for the analyst is to show that the selected cleaning agents or procedure actually cleans the surfaces in question. The drug is applied in solution form to the surfaces as in the recovery studies. The surfaces are then cleaned, rinsed, or treated as in the official cleaning procedure. Upon completion of cleaning, the surfaces are swabbed as before and if the cleaning has been successful the analysis will show the surfaces clean and free of analyte residue. The data in Table 7 describe the addition of each of the three cephalosporins in 5-10 mg amounts applied to surfaces in separate experiments, cleaned, and tested for residue as described above. It can be seen that this is a necessary step in the procedure in that it shows that the cleaning really works. [Pg.410]
