Sulfonylurea Dmgs

Six known and two new metabolites of glyburide, a sulfonylurea dmg applied in the treatment of insulin-independent Diabetes mellitus, were identified using DDA in LC-MS" on an ion-trap instmment [39]. DDA was also applied as an alternative to precursor-ion analysis in the screening and identification of moclobemide and remikiren metabolites [32]. More recently, ion-trap MS-MS in DDA mode was applied to perform the simultaneous metabolite identification and assessment of metabolic stability [40-41]... 

Relative potency alone does not determine dmg selection because maximal effectiveness is similar for all agents. A single daily dose of any sulfonylurea, except tolbutamide, is sometimes adequate to control blood glucose in NIDDM patients. 

Extensive reviews have been pubUshed, covering the Hterature to about 1967 (1 3). Pyrazoles and the benzopyrazoles have been well reviewed in References 4 and 5. More up-to-date reviews, though much narrower in scope, have been pubUshed on pyrazole oxides (6), dihydropyrazoles as insecticides (7), the anticancer dmgs anthrapyrazoles (8,9), and pyrazole sulfonylureas as herbicides (10). 

Adverse effects of the sulfonylureas are infrequent, occurring in about 4% of patients taking first-generation dmgs, and... 

Oral antidiabetic dmgs such as sulfonylureas are administered to patients who have Type 2 diabetes mellitus to stimulate beta cells to secrete insuMn. This results in an increase in insuMn cell receptors, enabling cells to bind to insuMn... 

Sulfonylureas are classified as first- and second-generation dmgs and each generation is divided into short-acting, intermediate-acting, and long-acting antidiabetics.
