The biosynthesis of pristinamycin 1 by Streptomyces pristinaespiralis is currently being studied in the RHONE POULENC laboratories. The natural amino acids threonine and proline have been shown to be incorporated directly into pristinamycin 1 and radiolabeled " C-pristinamycin I a has been prepared by incorporation of [U- C]proline. Interesting studies on the biosynthesis of virginiamycin Si described by Kingston and co-workers [44] showed that the phenylglycine residue was derived from phenylalanine via an intermolecular transfer of nitrogen. The biosynthesis of pristinamycin 11 by Streptomyces pristinaespiralis is currently being studied in the RHONE-POULENC laboratories. Previous studies [90, 91] on the biosynthesis of virginiamycin Mi( = pristinamycin Ilyv, see Table 7) by Streptomyces virginiae identified acetic acid, valine, glycine, proline and serine as the natural building blocks of virginiamycin Mj (see Fig. 10). Pristinamycin is a member of the streptogramin class of antibiotics effective against vancomycin-resistant S. aureus and vancomycin-resistant enterococcus. It is biosynthesized by the bacterium Streptomyces pristinaespiralis and consists of two components, pristinamycin lA (a macro-lide) and the depsipeptide pristinamycin HA (also known as streptogramin A) in the ratio of 30 70.
