Stomatitis everolimus

In an open phase II study of oral everolimus 10 mg/day for 28-day cycles in 35 patients with recurrent endometrial carcinoma, the most common drug-related adverse reactions were fatigue ( = 19), anemia (n —15), pain (n = 13), lymphopenia (n = 13), and nausea ( = 12) 17 required dosage reduction to 5 mg/day because of stomatitis ( = 10), hyperglycemia ( =3), or thrombocytopenia, hypertriglyceridemia, rash, and fever (one each) [35%... 

Mouth In four Japanese heart transplant recipients treated with everolimus, aphthous stomatitis necessitated dosage reductions in three and complete withdrawal in the other after a switch from mycophenolate mofetil [43 ]. Everolimus trough concentrations had not exceeded the target range at any time. They all recovered after dosage reduction or withdrawal. Aphthous stomatitis has been reported as an adverse reaction to sirolimus.

In a phase 1 trial, 20 patients with advanced RCC were treated with everolimus and sunitinib grade 3 toxicities noted were herpes stomatitis, leucopenia, vomiting. Streptococcus viridans endocarditis, gastrointestinal haemorrhage, mucositis, febrile neutropenia and thrombocytopenia with bleeding. Grade 4 toxicities were anaemia and pulmonary embolism [141. 

In a phase 2 study of everolimus monotherapy in patients with nonfunctioning neuroendocrine tumours or pheochromocytomas/paragangliomas, the major grade 3/4 adverse events were thrombocytopenia (14.7%), hyperglycaemia (5.9%), stomatitis (5.9%) and anaemia (5.9%) [16 ].
