Solubilization progesterone

In a third part, we compare the ability of both compounds to solubilize progesterone (a hydrophobic steroid hormone virtually insoluble in water) and to release the trapped progesterone suddenly and at constant pH. Possible biomedical uses of such systems will be commented.

The effect of alphadalone acetate on the solubility of alphaxolone is a phenomenon that remains to be explained. Simultaneous solubilization of steroid hormones has only been studied by Lbvgren and co-workers [168,169]. Oestradiol was solubilized independently of the C21 steroids and testosterone studied, i.e. the capacity for oestradiol was unaffected by the solubilization of the latter. However, the solubilization of ethinyl oestradiol with progesterone and with testosterone was dependent on the presence of the other. The solubility of 11 a-hydroxyprogesterone was enhanced by ethinyl oestradiol-an effect akin to that of alphaxolone and alphadalone acetate. In several other pairs of steroids solubilization was reduced. The amount of encapsulated model drug (retinyl acetate or progesterone) per mg of dried LS was determined through solubilization of LS in ethyl acetate at 60°C. Following filtration, the solution was analyzed by reverse-phase high-performance liquid chromatography (HPLC) to find the drug content. 

In a study of the solubilization of selected steroids by lysophosphatidylcholine, Saunders and co-workers [155, 156] showed that monopolar steroids such as cholesterol are much more readily solubilized than dipolar steroids such as progesterone, prednisolone, testosterone, oestrone, hydrocortisone and prednisone.
