In order to achieve maximum value to the Department, the immunoassays we develop will be validated through testing to determine within- and between-laboratory variability. At present, we plan to defer validation by the Association of Official Analytical Chemists (AQAC) until we have an assay which is more specific than the current atrazine assay, vrtuch is 40% cross-reactive to simazine. 

Triazines such as atrazine, propazine or simazine, are widely used herbicides. Currently, the determination of atrazine in water and soil samples is mainly done by GLC (1,2) or HPLC (3-5). However, these procedures require cumbersome cleanup steps which could be avoided by using immunoassays as an alternative approach to residue analysis. The sum of some other triazines that show cross reactivity (CR) in this form of ELISA (mAb - monoclonal antibodies, AM5D1), such as simazine, desethylatrazine and terbutylazine determined by the reference methods in RM08 adds up to a total of 2.3 dg L 1. Taking the CR of the monoclonal antibodies in the ELISA into account, however, the immunoassay should have given the value 0.48 jug L-1 and not 0.98 dg L-1 referring to about 20% of the sum of triazines present.
