Sequences joining

Workers in the early 1970s recognized that restriction enzymes provided tools not only for DNA mapping but also for constmction of new DNA species not found in nature. A collection of recombinant DNA species consisting of many passenger sequences joined to identical vector molecules is called a hbrary. Individual recombinant DNAs are isolated from single clones of the Hbrary for detailed analysis and manipulation.

Intracatenary cross-linked macromolecules occur very often in nature. With the enzyme, ribonuclease, for example, the single peptide chain is cross-linked to itself via four disulfide bridges (Figure 2-5). Another protein, insulin, has, on the other hand, two chains, A and B, of differing composition and peptide residue sequence, joined together by a total of two disulfide bridges.

We use the fact that our entire speech corpus is composed of large amounts of naturally occurring perfect joins , as every single example of two frames found side by side is an example of such.
