In pharmaceutical formulations, the most commonly used counterions are sodium and chloride ions. While no other reversed-phase column can retain or separate them, the Acclaim Trinity PI column overcomes this challenge by providing excellent separations and peak shapes for both analytes (see Figure 6.89). With the introduction of Acclaim Trinity PI, a broad range of hydrophobic and hydrophilic APIs that possess either acidic or basic functional groups can be separated along with their respective positive or negative counterions. As an example, the separation of the hydrophilic and basic API, 1,1-dimethylbiguanide (metformin), from its chloride counterion can be achieved using a mobile phase with a relatively low amount of acetonitrile (20% v/v) under isocratic conditions. [Pg.1316]
