4 Selection of phage libraries on purified, immobilized antigen 

Selection of phage antibody libraries by panning on immunotubes 

Rinse the tube three times with PBS and block the tube by filling it to the brim with PBS contaming 3% (w/v) skimmed milk powder and incubating for 1-2 h at 37 C If the antigen is tmstable at 37 °C block at room temperature for 2 h. 

Rinse the tube by completely filling it ten times in PBS containing 0.1% Tween 20, then ten times in PBS using a wash bottle. 

Elute the phage which have bound to the tube by adding 1 ml of freshly made 100 mM triethylamine and incubating at room temperature for 10 min.
