Drawbacks of the RTP ports are that they cannot be hooked into the isolator clean in place (CIP) system so, they must always be considered dirty until an adequate cleaning procedure is developed. There are also size limitations to the RTP ports and their corresponding containers. Once a certain size port is selected for the isolator, this is the only diameter container that can be used, unless the isolator port is modified. The port also requires an additional penetration of the isolator wall and serves as another potential point for air leaks. In addition, the RTP containers are not amenable to the transport of flasks into and out of the isolator they require rotation and maneuvering of the RTP container to dock it into the isolator. Unless the port is mounted into the isolator floor, which will take up valuable space, the ability to transport liquids into and out of the isolator is limited to securely closed vials or bottles. The final limitation is the tendency of the seals on the mating surfaces of the RTP ports to become contaminated with potent material during transport operations. Procedures should be put in place to ensure that the mating surfaces of the RTP ports are thoroughly decontaminated as soon as the transport container is undocked from the isolator wall. 

The problem of securing crystalline digitalonic lactone is somewhat simpler than is the case with the parent sugar, provided one has Digi-talinum verum, a substance usually commercially available in normal times. The preparation and purification of the glycoside from digitalis seeds has been described by Kiliani. The same worker has provided a method for the isolation of digitalonic lactone which represents a substantial improvement over earlier procedures. 

Rework procedures are usually employed to upgrade final products and key intermediates, even if the identity of the troublesome impurity has not been secured. Resubjecting the product to the original crystallization and isolation conditions may not sufficiently remove the key impurity. Rework procedures may need to be developed based on chemical information and intuition. 

The standard operating procedure (SOP) manual contains the procedures validated by the laboratory it is a complete set of instructions for pre-analytical, analytical and post-analytical methodology and also procedures for quality assurance/control, chain-of-custody and security. Each step in the handling of the specimen should be evaluated, optimized where possible and documented in the SOP. Important steps in the analytical process include collection, transport and accessioning of the specimen, sample preparation, isolation and detection of the analytes, production of the report and disposal of the specimen. This chapter focuses on the quality assurance and control issues for analytical method development and validation as well as statistical representation of the data. 

Until about 20 years ago the only way in which an alkaloid isolated from cadaveric material could be identified was by means of color and crystal tests. Both of these date back for well over a century, the color test being the older of the two. It has the great advantage of speed and simplicity and is ideally suited to rapid screening procedures.
