Screening procedures crystallization

Finally, libraries aimed to chiral resolution of racemates will be covered here in particular, the use of chiral stationary phases (CSPs) has recently been reported for the identification of materials to be used for chiral separation of racemates by HPLC. The group of Frechet reported the selection of two macroporous poly methacrylate-supported 4-aryl-1,4-dihydropyrimidines (DHPs) as CSPs for the separation of amino acid, anti-inflammatory drugs, and DHP racemates from an 140-member discrete DHP library (214,215) as well as a deconvolutive approach for the identification of the best selector phase from a 36-member pool library of macroporous polymethacrylate-grafted amino acid anilides (216,217). Welch and co-workers (218,219) reported the selection of the best CSP for the separation of a racemic amino acid amide from a 50-member discrete dipeptide iV-3,5-dinitrobenzoyl amide hbrary and the follow-up, focused 71-member library (220). Wang and Li (221) reported the synthesis and the Circular Dichroism- (CD) based screening of a 16-member library of CSPs for the HPLC resolution of a leucine ester. Welch et al. recentiy reviewed the field of combinatorial libraries for the discovery of novel CSPs (222). Dyer et al. (223) reported an automated synthetic and screening procedure based on Differential Scanning Calorimetry (DSC) for the selection of chiral diastereomeric salts to resolve racemic mixtures by crystallization. Until about 20 years ago the only way in which an alkaloid isolated from cadaveric material could be identified was by means of color and crystal tests. Both of these date back for well over a century, the color test being the older of the two. It has the great advantage of speed and simplicity and is ideally suited to rapid screening procedures. It has recently secured a new lease on life owing to the way in which it may be used for locating compounds on paper and thin-layer chromatograms. 

It seems clear that while medical applications of liquid crystal thermography are extensive and imaginative, the technique is most useful for screening procedures, and there are only a few cases where it has proven diagnostic capability. 

The key Pd-catalyzed coupling step to the indole was optimized by judicious choice of the protection group on 3-butyn-l-ol, screening of different reaction parameters such as bases and additives, and implementing a fine-tuned, yet practical work-up procedure. Tryptophol 27 was isolated in 76-82% yield by crystallization. 

Antibodies produced by this procedure were screened for their ability to react with the hapten to form the vinylogous amide 6, which has a convenient UV chromophore near 318nm, clear of the main protein absorption. Two antibodies selected in this way catalysed the expected aldol reaction of acetone with aldehyde 7 by way of the enamine 8 (Scheme 3) the remainder did not. These two effective aldolase mimics have been studied in some detail, and a crystal structure is available for (a Fab fragment of) one of them.126,281...
