Fixation for Enzyme Cytochemistry Using Microwave at Relatively [Pg.64]

The microwave processor is fitted with a thermometer, timer, stirrer, and power level controller (150 W to 400 W) (MI-77, Azumaya Company, Tokyo, Japan). One glass Petri dish with 70-ml capacity containing 50 ml of a mixture of 2% paraformaldehyde and 0.5% glutaraldehyde containing 45 mg/ml sucrose in 0.1 M PBS (pH 7.4) is placed in a large Petri dish containing 250 ml of chilled water in the processor. The specimens are heated in a microwave oven for 10 min at 4°C at a low energy level of 150 W. [Pg.64]
