Spin the isolated nuclei onto poly(L-lysine)-coated silicon chips, using a minibench centrifuge with a 1.5-ml Eppendorf tube half-filled with polymerized resin (used to support the silicon chip). Overlay each silicon chip with 0.5 ml of the isolated nuclear suspension and spin for 5 min at 100 g. Poly(L-lysine) (MW 271,000 Sigma, UK) is dissolved in distilled water at 1 mg/ml, and a 50-/il drop is placed on each silicon chip and left for 45 min to adsorb to the surface. After a brief rinse, the chips can be used immediately, or kept for up to 2 weeks in the refrigerator. [Pg.132]
