The replica-plating method may be used to select many types of mutants for which it would be tedious and inefficient to screen individually. The more common application is to isolate auxotrophic, radiation-sensitive, and conditional-lethal mutants. The so-called conditional-lethal mutants are those which, because they are defective in some manner, survive and grow under permissive conditions but perish under restrictive conditions. [Pg.135]

A replica-plating method for assessment of the bacterial composition of cotton dust. In Proc. Ninth Cotton Dust Res. Conf., pp. 63-66 (P.J. Wakelyn and R.R. Jacobs, eds.). National Cotton Council, Memphis, TN. [Pg.32]

The dotted circles represent colonies that do not grow on tetracycline. All the DNA sequences (or their complements) that encode the selected peptide sequence are synthesized by the solid-phase method and made radioactive by phosphorylating their 5 ends with from [ P]-ATP. The replica plate is exposed to a mixture containing all these probes and au-toradiographed to identify clones with a complementary DNA sequence. Positive clones are then sequenced to determine which ones have a sequence matching that of the protein of interest. Some of them may contain the desired gene or a significant segment of it. [Pg.156]

Immunological methods may be employed to detect the products of the foreign gene. After transformation, growth and replica-plating, the... [Pg.237]

This is an Academic Press Replica Reprint reproduced directly from the pages of a title for which type, plates, or film no longer exist. Although not up to the standards of the original, this method of reproduction makes it possible to provide copies of books which otherwise would be out of print. [Pg.379]

Cottin (C50) earlier studied pastes of Ciment Fondu, largely by the TEM replica method. The microstructures of pastes of low w/c ratios hydrated for 7 days at 12°C were too compact to show more than occasional detail, but with pastes of w/c = 0,7 much detail could be observed in cavities exposed by fracture, which were lined with small crystals of CAH,o and sometimes contained plates of C,AHg. The crystals of CAH[q, which were hexagonal prisms up to 1 pm long and 0.2 pm wide, were present in rounded aggregates. Hydrous alumina was seen in a paste of w/c = 1.0 hydrated at 30"C it occurred in the lining of a cavity, mainly as rounded masses of randomly oriented platelets. [Pg.326]
