Figure 17. (A) The preparation of an electrically wired enzyme by the reconstitution technique, involving the removal of the native FAD cofactor from the enzyme (e.g., GOx) and the incorporation of the artificial FAD-ferrocene dyad into the apo-enzyme. (B) Cyclic voltammograms of a system consisting of ferrocene-FAD-reconstituted GOx (1.75 mg mL ) at various concentrations of glucose (a) 0, (b) 1, (c) 3 and (d) 20.5 mM. Experiments were performed in 0.1 M phosphate buffer, pH 7.3, at 35 C, using a cystamine-modified Au electrode, potential scan rate 2 mV s , under argon. Inset calibration curve of the biocatalytic current (0.5 V vs. SCE) at different glucose concentrations. Figure 17. NMR study of Mb revealed that there are two interconverting protein forms in solution at equilibrium, which differ in the orientation of the heme by a 180° rotation about the 5,15-meso axis (Fig. 19). - The dominant form possesses the same heme orientation as found in the single crystal (A of Fig. 19) and is called the major form. The protein with the reversed heme orientation (B of Fig. 19) is called the minor form. Although the concentration of the minor form relative to the major form is small in native Mb, it can be raised using the reconstitution technique. The reconstitution of apoprotein with heme initially... [Pg.221]

Reconstitution techniques to machine specimens from pre-tested broken specimens make it possible to generate new surveillance test data. Charpy impact specimens and fracture toughness test specimens can be reconstituted by methods such as the ASTM E1753, Standard Guide for Reconstitution of Irradiated Charpy-sized Specimens, and JEAC 4201, Method of Surveillance Tests for Structural Materials of Nuclear Reactors. [Pg.102]
