Quinethazon

Chloro-6-sulfonamido-4(3 f)quinazo]inones are more effective than the mercury diuretics and can be administered orally. The 1,2-dihydro derivatives are even more effective and one (59) has been marketed under the name "Quinethazone. ... [Pg.306]

This drug bears some formal chemical resemblance to the benzothiatriazines referred to above [327] in that a nitrogen atom occupies the position corresponding to C3 in the thiazide series. (XXVll, Figure 1.1) Clopamide (Aquex, Brinaldix) has obvious features in common with quinethazone and chlorexolone for example and can be regarded either as a piperidinocarboxamide or a substituted carboxylic hydrazide of the familiar o-chlorobenzenesulphonamide. [Pg.43]

The thiazidelike compounds, including chlorthalidone Hygrown), quinethazone (Hydromox), and metolazone (Zaroxolyn) have similar mechanisms of action, but they differ substantially from one another in their duration of action, the degree of carbonic anhydrase inhibition, and the dose required for maximum natriuretic activity. [Pg.245]

Thiazide diuretics consist of two distinct groups those containing a benzothiadiazine ring, such as hydrochlorothiazide and chlorothiazide, referred to as thiazide diuretics, and those that lack this heterocyclic structure but contain an unsubstituted sulfonamide group. The latter are called thiazidelike diuretics they include metolazone, xipamide, and indapamide. The major thiazide and thiazidelike drugs available in the United States are bendrofiumethiazide, benzthiazide, chlorothiazide, hydrochlorothiazide, hydroflumethiazide, methyclothiazide, polythiazide, and trichlormethiazide and chlorthalidone, indapamide, metolazone, and quinethazone, respectively. [Pg.245]

Quinethazone 1169 and metolazone 1170 are used medically as diuretics, and alfazosin 1171, an cti-adreno-receptor antagonist related to prazosin is used to improve urinary flow rate in the treatment of benign prostatic hyperplasia. [Pg.249]

The discovery that the cyclization of one of the sulfonamide groups and the adjacent amine in classic monocyclic diuretics leads to an increase in potency is discussed in the next section. A similar increase in potency is also observed in the anthranilic acid based diuretics. Thus the fusion of anthranilic acid (87-1), which may show diuretic activity in its own right, with propionamide leads to the ring closure and formation of (87-2). Reduction of the unsaturation in the heterocyclic ring then affords the diuretic quinethazone (87-3) [97]. [Pg.484]
