Studies are being performed to determine the effect of presystemic and hepatic first-pass metabolism on the toxicity and carcinogenicity of xenobiotics. For a nontherapeutic toxic substance, the existence of a first-pass effect is desirable, because the liver can bioinactivate it, preventing its distribution to other parts of the body. On the other hand, first-pass metabolism may increase its toxicity by biotransforming the toxicant to a more toxic metabolite, which can reenter the blood and exert its toxic effect. [Pg.475]
