In context with structure-function analyses of operational photosynthetic complexes so-called inside-out vesicles from thylakoids of A. tabacum var. JWB were used [14, 15]. These vesicles have the advantage to originate from the partition region of the thylakoid membrane and to occur as particles in which by vesicle fonnation a defined membrane side, namely the originally towards the lumen directed surface becomes surface exposed. This is obtained by an isolation procedure in which distribution between the two-phase gradient polyethylene glycol dextran is used [15J. These inside-out vesicles represent an ideal material for immunological analyses of structure-function relationships. In a first paper we report here on the reaction of monospecific antisera towards the main components of lipids and xanthophylls with these vesicles.
