Pharmaceutical medicine industrial roles

The new discipline of pharmaceutical medicine is a speciality which has only very recently become recognized in its own right as a speciality within medicine. Indeed, the Faculty of Pharmaceutical Medicine of the Royal Colleges of Physicians was only founded in 1989 in the United Kingdom and the Academy in the United States even more recently in 1993. Like many new ventures, this new medical speciality is not seen by all today as one of the premiere medical roles. Today, multi-parallel synthesis lies at the forefront of organic and medicinal chemistry, and plays a major role in lead discovery and lead optimization programs in the pharmaceutical industry. The first solid-phase domino reactions were developed by Tietze and coworkers [6] using a domino Knoevenagel/hetero-Diels-Alder and a domino Knoevenagel/ene protocol. Reaction of solid-phase bound 1,3-dicarbonyl compounds such as 10-22 with aldehydes and enol ethers in the presence of piperidinium acetate led to the 1-oxa-1,3-butadiene 10-23, which underwent an intermolecular hetero-Diels-Alder reaction with the enol ethers to give the resin-bound products 10-24. Solvolysis with NaOMe afforded the desired dihydro-pyranes, 10-25 with over 90 % purity. Ene reactions have also been performed in a similar manner [7]. [Pg.569]

The roles of the Pharmaceutical Industry and the FDA, and the relationships between them, have evolved considerably since the 1962 effectiveness amendments to the FD C Act, and today the two parties are engaged in a close, complex, generally supportive - if sometimes fractious -relationship. There is no doubt that the achievements of these two parties, both individually and together, have been spectacular - even historic -as their effectiveness and safety standards have been embraced by the whole world. Indeed, these standards are now recognised as a landmark in post-enlightenment science, as well as a forerunner of the current philosophy of evidence-based medicine. [Pg.613]
