Periodate Oxidation Followed by Reductive Amination

Coupling via Periodate Oxidation Followed by Reductive Amination 

The following generalized method is based on the procedure described by Heath et al. (1981) for the coupling of immunoglobulins to liposomes containing gly-cosphingolipids. 

Periodate-oxidize a liposome suspension containing glycolipid components according to Section 2. Adjust the concentration of total lipid to about 5 mg/ml. 

5 ml of protein solution to each milliliter of liposome suspension with stirring. 

Incubate for 2 h at room temperature to form Schiff base interaaions between the aldehydes on the vesicles and the amines on the protein molecules. 

Dissolve the protein to be coupled in 20mM sodium borate, 0.15M NaCl, pH 8.4, at a concentration of at least lOmg/ml.
