Coupling via Periodate Oxidation Followed by Reductive Amination [Pg.562]

The following generalized method is based on the procedure described by Heath et al. (1981) for the coupling of immunoglobulins to liposomes containing gly-cosphingolipids. [Pg.562]

Periodate-oxidize a liposome suspension containing glycolipid components according to Section 2. Adjust the concentration of total lipid to about 5 mg/ml. [Pg.562]

5 ml of protein solution to each milliliter of liposome suspension with stirring. [Pg.564]

Incubate for 2 h at room temperature to form Schiff base interaaions between the aldehydes on the vesicles and the amines on the protein molecules. [Pg.564]

Dissolve the protein to be coupled in 20mM sodium borate, 0.15M NaCl, pH 8.4, at a concentration of at least lOmg/ml. [Pg.893]
