Periodate-lysine-paraformaldehyde

Fix tissue with periodate-lysine-paraformaldehyde (PLP) fixative. [Pg.299]

McLean IW, Nakane PK (1974) Periodate-lysine-paraformaldehyde fixative A new fixative for immunoelectron microscopy. J Histochem Cytochem 22 1077-1083... [Pg.60]

Periodate-lysine-paraformaldehyde (PLP) - It is used to increase cross-linking of molecules in the tissue to provide better fixation. Periodate oxidizes sugars attached to lipids and proteins and generates aldehydes, which bind lysine. Paraformaldehyde then cross-links the lysine. It uses 2% paraformaldehyde,... [Pg.21]

Kidney tissue is fixed with paraformaldehyde-lysine-periodate by vascular perfusion (Brown et al., 1996). Tissue slices are further fixed overnight at4°C with the same fixative and stored in PBS (pH 7.4) containing 0.02% sodium azide. They are placed in 30% sucrose in PBS for at least 1 hr, and then surrounded by a drop of Tissue-Tek embedding medium on a cryostat chuck before freezing by immersion in liquid nitrogen. Cryostat sections about 5 p,m thick are cut at a chamber temperature of -25°C, collected on Fisher Superfrost Plus charged slides, and stored at —20°C until use. [Pg.149]

Paraformaldehyde/Lysine/Periodate Fixative (PLP ) (2) (see Note 6) Place 30 mL of lysine stock in a 50 mL screw cap tube. Add 10 mL 8% paraformaldehyde. Add 0.085 g of Sodium w-periodate powder. Shake until powder dissolves. [Pg.57]
