Bromocriptine (Parlodel ) Activate postsynaptic D2 and blocks D1 DA receptors Start with 1.25 mg daily at bedtime, then 1.25 mg twice daily on week 2, increase to 2.5 mg twice daily, then increase by 2.5 mg daily every 2-4 weeks up to 15-45 mg daily divided 2-3 times daily...

Bromocriptine mesilate is 2-bromo-a-ergocryptine methane-sulphonate or 2-bromo-12 -hydroxy-2,-(l-methylethyl)-5 -(2-methylpropyl-5 a-ergotaman-3, 6, 18-trione methanesulphonate or Bromocriptinum (INN). It is the active ingredient in Parlodel dosage forms. 

Bromocriptine mesilate is marketed as Parlodel capsules (10 mg for the treatment of Parkinson s disease) and tablets (2.5 mg as lactation suppressor). Both forms have proved stable at least for 4 years when stored at ambient temperature in amber glass bottles (22). ... 

Bromocriptine mesilate in Parlodel tablets may be assayed in a non-specific way by direct uv-spectrophotometry following extraction with methanol (32). 

Dopamine-Boosting Medications. Levodopa/carbidopa (Sinemet), bromocriptine (Parlodel), pramipexole (Mirapex), and ropinirole (Requip) increase dopamine nenrotransmission in the brain by one or another mechanism. These medications do not reliably induce sleep, and in some patients are activating. They are certainly not true sedative-hypnotics. Bromocriptine, a dopaminomimetic that is a dopamine D2 receptor agonist, possesses expressed antiparkinsonian activity. It is used for treating all phases of idiopathic and post-encephalic Parkinsonism. However, it has a number of undesirable side effects, even causing mental disturbances in long-term use. The most common synonyms are parlodel, bromergon, and others.

Bromocriptine is generally taken after the evening meal at the initial dose of 1.25 mg the dose is then increased as tolerated. Most patients require 2.5-7.5 mg daily acromegalics require higher doses, up to 20 mg/d. Bromocriptine tablets may be administered intravaginally to reduce nausea.
