The 5-HT3 antagonist palonosetron is the first 5-HT3 antagonist to be approved for prevention of both acute and delayed CINV.40 Compared to the other 5-HT3 antagonists, palonosetron has a longer serum half-life (40 hours compared to 4 to 9 hours) and a higher receptor binding affinity, which may contribute to its efficacy in preventing delayed CINV.41... [Pg.303]

Administration Infuse IV over 30 seconds. Do not mix palonosetron with other drugs. Flush the infusion line with normal saline before and after administration of palonosetron. [Pg.1002]

Alosetron is a 5-HT3 antagonist that has been approved for the treatment of patients with severe IBS with diarrhea (Figure 62-5). Four other 5-HT3 antagonists (ondansetron, granisetron, dolasetron, and palonosetron) have been approved for the prevention and treatment of nausea and vomiting (see Antiemetics) however, their efficacy in the treatment of IBS has not been determined. The differences between these 5-HT3 antagonists that determine their pharmacodynamic effects have not been well studied. [Pg.1321]

Ondansetron is one of a group of 5HT3-receptor antagonists currently available in the UK, which also includes granisetron, tropisetron, dolasetron and palonosetron. Dexamethasone is a corticosteroid. Ranitidine is a histamine-2 (H2)-receptor antagonist. [Pg.185]

One of the major side-effects of cytotoxic chemotherapy is nausea and vomiting. It is essential therefore to control this with prophylactic antiemetic therapy. A number of agents are currently available and are effective at controlling chemotherapy-induced emesis. The most potent of these are 5HT3-receptor antagonists that include ondansetron, granisetron and palonosetron. [Pg.208]
