This enzyme [EC 3.6.1.6] catalyzes the hydrolysis of a nucleoside diphosphate to produce a nucleotide (that is, a nucleoside monophosphate) and orthophosphate. NDP substrates include IDP, GDP, UDP, as well as d-ribose 5-diphosphate.

Walseth et al. (W3) separated 5 -ribonucleoside monophosphates using ion-pair chromatography. The chromatognq>hic system has been used to analyze tissue nucleotides, to purify nucleoside monophosphates and cyclic nucleotides, and to monitor enzymatic reactions involving cyclic nucleotide phosphodiesterase, 5 -nucleotidase, and adenylate cyclase.

Synthesis of a phosphodiester bond in nucleic acids requires energy input. As a result, the nucleoside monophosphates in nucleic acids are built up from hydrolysis of nucleoside triphosphates. Cleaving a pyrophosphate from a nucleoside triphosphate yields a nucleoside monophosphate and enough free energy to make the formation of polynucleoside monophosphates (i.e., polynucleotides) thermodynamically favorable. 

It is apparent from the above considerations that an accelerated rate of purine biosynthesis de novo can occur, therefore, as a result of either elevated levels of PRPP in the cell or decreased levels of purine nucleotides especially the purine nucleoside monophosphates and ADP. Indeed, it seems likely that in many, if not all, clinical conditions to be described, alterations in the levels of both PRPP and purine nucleotides contribute to the accelerated rate of purine biosynthesis. The enzymatic machinery of the tricarboxylic acid cycle is located in a mitochondrial membrane system as mentioned above, the presence of nucleoside monophosphate and diphosphate kinases in this cell compartment enables the substrate-level phosphorylation associated with a-keto-glutarate oxidation to be coupled with the adenosine phosphates.
