SR the enzyme can be purified from erythrocytes [20] or as recombinant enzyme from bacteria [9,21,22]. Preferentially, we expressed the rat SR-cDNA as N-terminally poly histidine (His6)-tagged protein in bacteria and purified it upon applying a commercially available nickel-nitrilotriacetic acid affinity column (Neuheiser, Blau and Thony, unpublished). The noncleaved, highly active fusion protein is stored in liquid nitrogen at a concentration of 1 mg/ml and an activity of approximately 1 U/ml. 

Here, the specific and strong chelating interaction between 6 x Histidine and the tetradentate nitrilotriacetic acid (NTA) mediated by Ni2+ [43] was used for the immobilization of a reconstituted LHCII mutant, whose c-terminus was extended by 6 x Histidine residues via genetic engineering (cf. Fig. 13(A)). NTA terminated thiols and OEG (spacer) thiols were used to built up a mixed SAM (X = 0.4) on a substrate coated by 23 nm silver and 5 nm Au (the gold layer was used to protect the silver film from being oxidized).

Besides calcium minerals, the growth of a few other inorganic salts on solid surfaces has also been studied. Silica is another important biomineral [81, 182], yet it has not been studied extensively in the context of surface mineralization. Tahir et al. synthesized a new nitrilotriacetic acid (NTA)-terminated alkenethiol, which can self-assemble on gold surfaces. Using the well-known coordination of Ni + and NTA, the enzyme silicatein could be immobilized on the surface of gold nanoparticles. Exposure of these functional particles and control experiments in the absence of silicatein showed that silica precipitates only on silicatein-modified...
