Natural Product Target Elucidation

Camptothecin was discovered as an active anticancer drug isolated from the bark of Camptotheca acuminata. The anticancer activity of camptothecin was discovered in the 1960s by the National Cancer Institute (NCI) as part of a systematic effort to screen for novel anticancer agents derived from natural products. Monroe Wall and Mansuhk Wani identified the chemical structure of camptothecin. They also identified the chemical structure of taxol, again under the auspices of the NCI. Susan Hoiwitz was contracted by the NCI to elucidate the anticancer mechanisms of camptothecin. She found in the early 1970s that camptothecin induced DNA breaks and attested DNA and RNA synthesis. However, it is approximately 12 years later, only after DNA topo-isomerase I (Topi) had been identified in human cells, that Leroy Liu and his coworkers found that Topi was the cellular target of camptothecin [reviewed in [1]. [Pg.315]

Today, selected targets of interest are transferred to the HTS aiming at discovering a hit , and subsequently a lead structure within one to two months. After that time, the target concerned is replaced by a new one. Thus, rapid characterization and structure elucidation of the active principles of interest from natural sources are critical with reference to competition with synthetic libraries of pure and structurally defined compounds. Elaboration of LC-MS and LC-NMR techniques to accelerate structure elucidation of bioactive principles from natural sources is currently underway. Combination of these techniques with databases comprising a maximum of known natural compounds will probably contribute substantially to more interest in natural products for application in drug discovery. [Pg.143]

In the pharmaceutical industry, the techniques are being used to examine off-target effects particularly for the early identification of toxicity. MOA can be studied through metabolomics and can also be used as a quality control tool for complex mixtures such as foods or herbal medicines. Similarly, the tools and expertise of natural products chemists are essential in metabolomics, particularly in biomarker discovery (see also Volume 9). Biomarker discovery via untargeted metabolomics can lead to metabolite signatures (nuclear magnetic resonance (NMR) spectroscopy, mass spectrometry (MS), etc.) that are not present in current metabolomics databases. This is particularly true for plant secondary metabolism studies and nonmammalian metabolites. Structure elucidation then becomes critical to understanding the metabolomics results and for biomarker development. [Pg.596]
