AN EVALUATION OF FOUR MYCOLOGICAL MEDIA FOR ENUMERATION OF MOLD AMD YEAST IN GRAINS AND SEEDS... 

In each assay, 11 g of sample were combined with 99 ml of 0.1% peptone water which contained 0.01% tween 80. This mixture was shaken vigorously for two minutes. Serial dilutions (1 10) of samples were prepared, and 100 pi of each dilution were added to separate Petri dishes. Duplicate samples were surfaced plated in each previously-prepared, mycological media and dried at 26°C for one hour. 

Information presented in Tables 1 and 2 shows mold and yeast contaminations at the second and third day of incubation at 26 °C. Recovery of mold and yeast was significantly higher (P < 0.05) at the third day of incubation, than at the third day. The highest three-day recovery of mold (refer to Table 1) from the different mycological media was obtained with OGY agar, followed by DRBC, PCA, and SDA. 

Clinical specimens must be collected aseptically and then promptly transported to the clinical laboratory in a properly labeled sterile container. Specimens collected on swabs or transported to the clinical laboratory in a transport medium are unacceptable for mycological study. An adequate quantity of clinical material is necessary if the information obtained in the laboratory is to be meaningful. 

A novel application of the microbioassay was also developed for the discovery of compounds that inhibit Phytophthora spp. This protocol used the 96-well format for high-throughput capability and a standardized method for quantification of initial zoospore concentrations for maximum reproducibility. Zoospore suspensions were quantifiable between 0.7 and 1.5 x 105 zoospores/mL at an absorbance value of 620 nm. Subsequent growth of mycelia was monitored by measuring absorbance (620 nm) at 24-hour intervals for 96 hr. Full- and half-strength preparations of each of three media (V8 broth, Roswell Park Memorial Institute mycological broth, and mineral salts medium), and four zoospore concentrations (10, 100, 1000, and 10,000...
