Murine mAbs are antibodies of murine origin. The first murine mAb to be approved for clinical use was muromonab-CD3 (Orthoclone OKT3 ), an mAb with CD3 on Tcells as the molecular target, and used in organ transplantation. [Pg.53]

The first murine monoclonal antibody was approved for marketing in 1986/ when Orthoclone (CD3-specific antibody)/ or OKT3/ was approved. NoW/ humanized antibodies/ engineered so that HAMA response is mitigated have become mainstream therapy/ with such recent successes as Raptiva/ ErbituX/ and Avastin (Table 32.1). A successful antibody also needs to be potent and specific (6). The following sections describe how engineered antibodies can be produced to meet these requirements. [Pg.479]

Several humanized CD3 antibodies have been produced and tested, with the aim of reducing the adverse effects associated with the murine orthoclone antibody. The first results in renal transplant patients were encouraging, with promising clinical results, no evidence of antiglobulin response, and no or very few and minimal adverse effects attributable to various humanized monoclonal CD3 antibodies (16,17). In particular, careful patient monitoring failed to identify any significant cytokine release syndrome. [Pg.2397]
