Abuchowski et al. [28] prepared a reactive intermediate, activated PEGi (Fig- 3a). To cyanuric chloride (0.03 mol) dissolved in anhydrous benzene containing sodium carbonate was added monomethoxypolyethylene glycol (mPEG-OH with MW of 5000, 0.01 mol) and the mixture was stirred overnight at room temperature. The modifier thus prepared, activated PEGi, directly reacts with amino groups on the surface of protein molecule in an aqueous buffered solution at pH 9.2 and 4 C for 1 h (Scheme 3).
