As stated in the introduction, conceptual DFT is based on a series of reactivity descriptors mostly originating from a functional Taylor expansion of the E = E[N, v(r)] functional. These (<)nE/3NmSv(r)m ) quantities can be considered as response functions quantifying the response of a system for a given perturbation in N and/or v(r). In the case of molecular interactions (leading to a new constellation of covalent bonds or not), the perturbation is caused by the reaction partner. Rather than using an amino acid sequence to search SWISS-PROT, AACompI-dent of ExPASy Proteomic tools (http //www.expasy.ch/tools/) uses the amino acid composition of an unknown protein to identify known proteins of the same composition. The program requires the desired amino acid composition, the pi and molecular weight of the protein (if known), the appropriate taxonomic class, and any special keywords. The user must select from one of six amino acid constellations that influence how the analysis is performed. The Patterson synthesis (Patterson, 1935), or Patterson map as it is more commonly known, will be discussed in detail in the next chapter. It is important in conjunction with all of the methods above, except perhaps direct methods, but in theory it also offers a means of deducing a molecular structure directly from the intensity data alone. In practice, however, Patterson techniques can be used to solve an entire structure only if the structure contains very few atoms, three or four at most, though sometimes more, up to a dozen or so if the atoms are arranged in a unique motif such as a planar ring structure. Direct deconvolution of the Patterson map to solve even a very small macromolecule is impossible, and it provides no useful approach. Substructures within macromolecular crystals, such as heavy atom constellations (in isomorphous replacement) or constellations of anomalous scattered, however, are amenable to direct Patterson interpretation. Knowledge of the composition of fhe interstellar medium has been growing rapidly in the last 25 years. Spectroscopic techniques, including some that will be considered in Chapter 10, can sometimes reveal the "fingerprints" of molecules in the electromagnetic radiation received from interstellar space. In 1963 the existence of OH was reported, and in 1968 Charles Townes and coworkers identified the first polyatomic molecule, ammonia, in a cloud associated with the constellation Sagittarius. The next year water was identified, and Lewis Snyder found the first organic molecule, formaldehyde.
