Mobile phase compatible

Principles and Characteristics Multidimensional gas chromatography (MDGC) is widely used, due to the mobile-phase compatibility between the primary and secondary separating systems, which allows relatively simple coupling with less-complicated interfaces. In its simplest form, 2DGC can be carried out in the off-line mode. The most elementary procedure involves manual collection of effluent from a column, followed by reinjection into another column of a different selectivity (e.g. from an apolar to a polar column). Selecting proper GC-column combinations is critical. In on-line mode, the interface in MDGC must provide for the quantitative transfer of the effluent from one column... 

Typical mobile phases compatible with electrospray are methanol, acetonitrile, isopropanol, and volatile buffers. Many other solvents can also be used in ESI, including CHCI3 and THE. Although 100% water can be used, better sensitivity is obtained with some organic modifier... 

Module 3, Column and Mobile Phase Design (CMP). This is the core module for ECAT. It can currently specify i) analytical column and mobile phase constituents for reverse phase chromatography of common classes of organic molecules ii) reverse phase, ion exchange phase and hydrophobic interaction chromatography of proteins and peptides iii) a limited set of specialty classes of molecules best treated by straight phase chromatography (e.g., mono- and disaccharides). The rules for selection of the HPLC detector are under development within Module 3. Some of the rules for detector mobile phase compatibility are already encoded. A set of rules for detector selection is ready but not yet encoded. 

S.3 General Considerations for Buffers. The type of buffer that is chosen will depend on the wavelength of the method and the concentration of organic in the mobile phase. A judicious choice of type and concentration of buffer must be made to ensure mobile-phase compatibility. 

The separation of enantiomers is especially important in the pharmaceutical field, because drag enantiomers may produce different effects in the body. Enantiomer separations by chromatography require one of the components of the phase system to be chiral. This can be achieved by (a) the addition of a chiral compound to the mobile phase, which is then used in combination with a nonchiral stationary phase, or (b) the use of a chiral stationary phase in combination with a nonchiral mobile phase. The chiral phase can either be a solid support physically coated with a chiral stationary phase liquid or a chemically bonded chiral phase. For mobile-phase compatibility reasons, a chiral stationary phase is preferred in LC-MS. However, most chiral stationary phases have stringent demands with respect to mobile-phase compositiorr, which in turn may lead to compatibility problems.
