Microcrystalline cellulose, quantitation

The phase composition of glycine crystal forms during the drying step of a wet granulation process has been studied, and a model developed for the phase conversion reactions [88], X-ray powder diffraction was used for qualitative analysis, and near-infrared spectroscopy for quantitative analysis. The total cellulose (microcrystalline cellulose and carboxymethylcellulose sodium) quantitation is achievable through use of one of the following (1) total solid subtraction, (2) indirect quantification of sodium carboxymethylcellulose (NaCMC), or (3) direct quantification of the hydrolyzed monosaccharide. 

The solid phase in this case is particulate (e.g. microcrystalline cellulose) and the activity (complexed Ag E) is measured in the pellet after centrifugation. This activity is inversely proportional to the amount of antigen present in the sample since unreacted Ab (due to the absence of Ag) is quantitated. Although this method is more laborious than the first variant of sequential saturation, it has a wider applicability (Table 14.7B). These methods require highly purified antigens in sufficient amounts for labelling.

Siddiqi et al. (1995) separated alpha amino acids on tin(IV) selenoarsenate layers with a mobile phase of dimethyl sulfoxide (DMSO). Petrovic and Kastelan-Macan (1995) used scanning densitometry for the validation of quantitative amino acid analysis on mixed natural zeolite—microcrystalline cellulose layers. Ohtake et al. (1995) used cellulose TLC to analyze homocysteine from dried blood spots of patients with homocystinuria.
