The phenomena of association colloids in which the limiting structure of a lamellar micelle may be pictured as composed of a bimolecular leaflet are well known. The isolated existence of such a limiting structure as black lipid membranes (BLM) of about two molecules in thickness has been established. The bifacial tension (yh) on several BLM has been measured. Typical values lie slightly above zero to about 6 dynes per cm. The growth of the concept of the bimolecular leaflet membrane model with adsorbed protein monolayers is traceable to the initial experiments at the cell-solution interface. The results of interfacial tension measurements which were essential to the development of the paucimolecular membrane model are discussed in the light of the present bifacial tension data on BLM. [Pg.111]

Artificial biomembrane mimetic model systems are used to characterize peptide-membrane interactions using a wide range of methods. Herein, we present the use of selected membrane model systems to investigate peptide-membrane interactions. We describe methods for the preparation of various membrane mimetic media. Our applications will focus on small unilamellar vesicles (SUVs) and large unilamellar vesicles (LUVs) as well as on media more suited for nuclear magnetic resonance (NMR) techniques, micelles, and fast-tumbling two-component bilayered micelles (bicelles). [Pg.129]

Certain spectroscopic techniques, such as nuclear magnetic resonance (NMR) methods, require that the membrane mimetic, i.e., the lipid aggregate is not too large, and that the lipids exhibit a high degree of motion. For such studies, the micellar membrane model is often preferred. Micelles are relatively small (Fig. 3, top), which means that they rotate rapidly, on the time-scale required for NMR. Compared to phospholipid vesicles, it has been observed that peptides tend to be structured in micelles, and that SDS in particular seems to induce a high degree of a-helical secondary structure.
