Daytrana is a newly approved methylphenidate transdermal system. It is a 9-h adhesive matrix patch and comes in four sizes a 12.5-cm2 patch that has a delivery rate of l.lmg/h, an 18.75-cm2 patch that has a delivery rate of 1.6mg/h, a 25-cm2 patch that delivers 2.2mg/h, and a 37.5-cm2 patch that delivers 3.3mg/h.

Only few transdermal products (e.g., steroidal hormone, caffeine, theophylline, fentanyl, scopolamine, nicotine, methylphenidate) have been tested or marketed for use in the pediatric population. The development of transdermal products in pediatric doses could be very beneficial for children who are unable to tolerate oral medications. The need for several sizes of patches to cover different doses needed by different subsets of the pediatric population and to avoid accidents with cutting adult patches can be a limitation. The younger, the better permeation. Hence a compromise between topical versus transdermal efficacy and safety should be sought. 

The results are reported of an investigation into the stability, in-vitro drug delivery and adhesive properties of drug-in-adhesive transdermal drug delivery systems. These systems are composed of a flexible backing film, a fluoropolymer release liner and active adhesives, which vary in their monomeric compositions and functionalities. The adhesives are composed of an acrylic pressure-sensitive adhesive, an amine-compatible silicone pressure-sensitive adhesive and methylphenidate base, as the drug. 4 refs.

Methylphenidate is both available as the racemic mixture and its (R,R)-stereoisomer. It is used as the hydrochloride salt and preparations include immediate (i/r)- and modi-fied-release (m/r) formulations and transdermal patches.
